Passage through the cell cycle requires the successive activation of different cyclin‐dependent protein kinases (CDKs). These enzymes are controlled by transient associations with cyclin regulatory subunits, binding of inhibitory polypeptides and reversible phosphorylation reactions. To promote progression towards DNA replication, CDK/cyclin complexes phosphorylate proteins required for the activation of genes involved in DNA synthesis, as well as components of the DNA replication machinery. Subsequently, a different set of CDK/cyclin complexes triggers the phosphorylation of numerous proteins to promote the profound structural (...) reorganizations that accompany the entry of cells into mitosis. At present, much research is focused on elucidating the links between CDK/cyclin complexes and signal transduction pathways controlling cell growth, differentiation and death. In future, a better understanding of the cell cycle machinery and its deregulation during oncogenesis may provide novel opportunities for the diagnostic and therapeutic management of cancer and other proliferation‐related diseases. Following the publication of the seminal Fear of Freedom, Erich Fromm applied his unique vision to a critique of contemporary capitalism in The Sane Society. Where the former dealt with man's historic inability to come to terms with his sense of isolation, and the dangers to which this can lead, The Sane Society took his theories one step further. In doing so it established Fromm as one of the most controversial political thinkers of his generation. Anaylsing how individuals conform (...) to contemporary capitalist and patriarchal societies, the book was published to wide acclaim and even wider disapproval. It was a scathing indictment of modern capitalism and as such proved unwelcome to many. Unwelcome because much of what Fromm had to say was true. Today, as we settle into the challenges of the 21st century, Fromm's writings are just as relevant as when they were first written. Read it and decide for yourself - are you living in a sane society? Current fears of a “reproducibility crisis” have led researchers, sources of scientific funding, and the public to question both the efficacy and trustworthiness of science. Suggested policy changes have been focused on statistical problems, such as p-hacking, and issues of experimental design and execution. However, “reproducibility” is a broad concept that includes a number of issues. Furthermore, reproducibility failures occur even in fields such as mathematics or computer science that do not have statistical problems or issues with experimental design. Most (...) importantly, these proposed policy changes ignore a core feature of the process of scientific inquiry that occurs after reproducibility failures: the integration of conflicting observations and ideas into a coherent theory. This text is a short introduction to logic that was primarily used for accompanying an introductory course in Logic for Linguists held at the New University of Lisbon (UNL) in fall 2010. The main idea of this course was to give students the formal background and skills in order to later assess literature in logic, semantics, and related fields and perhaps even use logic on their own for the purpose of doing truth-conditional semantics. This course in logic does not replace (...) a proper introduction to semantics and is not intended as such, although parts of Chapter 1 and 4 could be used to supplement an introductory course in semantics. This paper makes the assumption that organ transplantation is, under some conditions at least, a proper use of communal medical resources. Proceeding from this assumption, the author: (1) sketches the history of the problem; (2) briefly examines the prevalent models of communal structure and offers an alternate version; (3) discusses notions of justice and obligation derived from these different models; (4) applies these to the practice of harvesting organs for transplantation; and then (5) offers a different process for harvesting organs (...) from the newly dead.If community is viewed as united by a set of shred goals and common values among which the value of community itself is important, then certain reciprocal obligations among members obtain. I suggest that routine salvage of organs from the newly dead be instituted but that it be routine salvage with a twist: rather early in life all members of the community are given the opportunity to refuse but their refusal carries the reciprocal condition that they cannot later become the recipients of that which they refuse to others. This paper examines the role of the physician in a pluralistic community. A personal and communal sense of identity must resolve a vast array of often conflicting backgrounds and contexts in order to function smoothly. Physicians are neither entitled to impose their own moral views on their patients nor expected to surrender their own moral agency. Several illustrative cases are given. The solution of inevitable conflicts is embodied within the context of the situation, but since irreconcilable differences remain, a resolution (...) is not always possible. I argue in this thesis that Hegel's work provides the basis for the categorization and evaluation of historical epochs which are distinct and separate cultures. In order to prove this contention I present an interpretation of Hegel's philosophy that yields a coherent account of the different aspects of Hegel's philosophy of culture. This is necessary considering the array of different views on Hegel's philosophy that exist. The principles of Hegel's philosophical and historical discussions are then employed to gain insight into (...) cultural and historical phenomena including post-Hegelian developments. ;The thesis first explores reasons for different readings of Hegel. This discussion is accompanied by an explanation of Hegelian terms and philosophical assumptions. I argue that Hegel's philosophy allows for the evaluation of different cultures, which is usually considered to be an impossible task due to the historical situation or cultural affiliation of any researcher. Furthermore, I maintain that this aspect of Hegelian philosophy makes it difficult to classify Hegel as a modern, a post-modern or even a post post-modern thinker. ;It is a well known fact that Hegel's categorization of history is marred by inconsistencies. However, this defect can be remedied using Hegel's own philosophical presuppositions. Consequently, the thesis first presents Hegel's ordering of historical epochs based on a literal reading of his Philosophy of History. Second, I develop an hypothetical ordering of history which is constructed in accordance with Hegel's philosophical outlook. The hypothetical historical model reveals that Hegel's Prussia does not constitute the stage of absolute knowing and that humanity is still on its way towards this stage. (shrink)

The Enigma of Modern Economics The effects of the present economic system are remarkably ambiguous. When we compare modern society with any preceding society in history it becomes evident that the ability to produce wealth is its distinguishing feature. It also is evident that the most highly productive and technologically advanced societies of the world (...) are at the root of grave social and ecological disturbances that are pushing humankind to the edge of global catastrophe. The scope and complexity of the adverse effects and risks, of the degradation to the natural surroundings and to social cohesion, are unprecedented in history. Although those phenomena are well known, they are not widely understood. The positive effects of modern economics are evident. The outstanding capacity to produce a vast array of technical instruments and rational organizations which serve as problem solving tools have opened the way to a level ofprosperity, and indeed of affluence, that is unparalleled in the history of civilization. In sharp contrast to this story of success is, however, the equally impressive record ofdestruction. The gulf between those who possess vast material means and those who possess the minimum for survival is widening both on a global scale and within individual affluent societies. The relation between increased productivity and ecological destruction seems to parallel that between growing wealth and growing human misery. The high rate of exploitation of natural resources and the artificial changes in climate are endangering the viability of future generations to the extent that the extinction of species and cultures is, finally, not a vague, futuristic threat, but a clear and present danger to life as we know it. The logical cumulative question is, then, Why is the current economic system both extraordinarily productive and destructive? Paramount efficiency in the production of wealth and success Never before has society been so efficient in accumulating wealth. This is the most striking feature of the upper financial strata of humanity, of, for example, the OCED countries. Whatever factor can be singled out, the formula remains the same: high levels of input of energy and other resources, and high output in terms of tools for production and consumer goods. For decades the general trend of income, savings, investments, and consumer expenditures, whatever their fluctuations, has been upward. In spite of the grievances and alleged cases of injustice, ever growing prosperity and affluence has become the normal expected result of the economic process. Wealth is produced not only in terms of achieved output, but also in terms of the capacity to tackle any difficulties through an expansion of the economic process. This may be the most important feature of present economic performance, even more important than its excellence in increasing the output of goods and services. Wealth is accumulated, moreover, not only in terms of material goods and services, but in terms of the options we have in reacting to the reality of needs and scarcities. The goal is to present how shared decision-making in paediatric oncology occurs from the viewpoints of parents and physicians. Eight Swiss Pediatric Oncology Group centres participated in this prospective study. The sample comprised a parent and physician of the minor patient. Surveys were statistically analysed by comparing physicians’ and parents’ perspectives and by evaluating factors associated with children’s actual involvement. Perspectives of ninety-one parents and twenty physicians were obtained for 151 children. Results indicate that for six aspects of information provision (...) examined, parents’ and physicians’ perceptions differed. Moreover, parents felt that the children were more competent to understand diagnosis and prognosis, assessed the disease of the children as worse, and reported higher satisfaction with decision-making on the part of the children. A patient’s age and gender predicted involvement. Older children and girls were more likely to be involved. In the decision-making process, parents held a less active role than they actually wanted. Physicians should take measures to ensure that provided information is understood correctly. Furthermore, they should work towards creating awareness for systematic differences between parents and physicians with respect to the perception of the child, the disease, and shared decision-making. We reconstruct recent work on macrosocial stress as if it were an instance of a research strategy that tests point-alternative hypotheses within a full-fledged research program. Because this strategy is free of various deficits that beset dominant strategies, our article demonstrates one way in which the confidence crisis may be overcome.

The prevalence and negative impact of brain disorders are increasing. Clinical Neuropsychology is a specialty dedicated to understanding brain-behavior relationships, applying such knowledge to the assessment of cognitive, affective, and behavioral functioning associated with brain disorders, and designing and implementing effective treatments. The need for services goes beyond neurological diseases and has increased in areas of neurodevelopmental and psychiatric conditions, among others. In Europe, a great deal of variability exists in the education and training of Clinical Neuropsychologists. Training models include (...) master’s programs, continuing education courses, doctoral programs, and/or post-doctoral specialization depending on the country, with no common framework of requirements, although patients’ needs demand equal competencies across Europe. In the past five years, the Standing Committee on Clinical Neuropsychology of the European Federation of Psychologists’ Association has conducted a series of surveys and interviews with experts in the field representing 30 European countries. The information, along with information from the existing literature, is used in presenting an overview of current and relevant topics related to policy and guidelines in the training and competencies in Clinical Neuropsychology. An option for the way forward is the EuroPsy Specialist Certificate which is currently offered in Work- and Organizational psychology and Psychotherapy. It builds upon the Basic Certificate and complements national standards without overriding them. General principles can be found that can set the basis for a common, solid, and comprehensive specialty education/training, sharpening the Neuropsychologists’ competencies across Europe. The requirements in Clinical Neuropsychology should be comparable to those for the existing specialty areas in the EuroPsy model. Despite the perceived challenges, developing a Specialist Certificate appears a step forward for the development of Clinical Neuropsychology. Recommendations are proposed towards a shared framework of competencies by the means of a common level of education/training for the professionals in Europe. Benchmarking training standards and competencies across Europe has the potential of providing protection against unqualified and ethically questionable practice, creating transparency, raising the general European standard, and promoting mobility of both Clinical Neuropsychologists and patients in Europe, for the benefit of the professional field and the population. Mr. Mike tells disturbing, pointless stories, not to comfort children, but to make them aware of the darkness of the human condition. Most of Michael O'Donoghue's work was the darkest of dark comedy. O'Donoghue captures common mortality/vulnerability in a series of sketches in which he appears (...) as an “impressionist.” Michael O'Donoghue was sometimes called “the Mayor of Death,” and the confrontation with death is a key aspect of existentialist philosophy. For existentialists, mortality is one of the defining characteristics of human existence. Empirical tests of Schwartz’s theory of culture-level value priorities have predominantly been performed using an averaging approach–as values of the average individual in a culture. However, from a theory of measurement standpoint such an approach seems inadequate. We argue that the averaging approach is an insufficiently accurate methodology in capturing the compatibilities-incompatibilities between values of individuals within cultures. We propose an approach based on the distribution of values of individuals in a given culture–the distribution approach. Using data from two rounds (...) of the European Social Survey, we show how frequencies of specific individual value priorities in a culture can be used towards the description of culture-level value preferences. We recommend a re-conceptualization of Schwartz’s culture-level value theory to an orthogonal two-dimensional structure, namely as Preservation vs. Alteration and Dominance vs. Amenability, which we explain based on heterogeneity in socioecological indicators across countries. We conclude that societal challenges may influence the cultural value climate across countries. (shrink)

[The Hermeneutics and Non-Dogmatic Dialectics of Gadamer].

This paper briefly reviews the papers in this special section of HCA and makes the point—a point which should be obvious—that statistics are useful only as guidelines but tell one nothing about the individual patient in front of you. Chronological age merely shows what is true of most but decidedly not of all patients in a particular age group. To ration on the basis of age alone is unfair to the individual denied treatment and damaging to the community because it (...) disturbs the solidarity which comes about because most members of the community feel that the community has obligations beyond those of not directly harming them; indeed, what produces solidarity is the feeling that members of a community will do their best to come to each others help. Rationing on the basis of age alone denies people of equal treatment under the law and—when it comes to the elderly—is a type of age discrimination. It is pointed out that what matters is a patient’s disease and not his/her age. A permanently vegetative person 8 years of age is a much sadder occurrence than it would be at age 90—but the critical fact is that both are permanently vegetative. Age cuts both ways—it is irrational to spend hundreds of thousands of dollars, untold amounts of time, energy and devotion to the 520 gm infant with a gr IV diffuse haemorrhage whose chance of leading a sentient life is close to zero and to hesitate before using a diagnostic MRI on a patient who is 90 but fully alert and enjoying life. It is concluded that age as an independent variable in the allocation of resources is ethically highly problematic. (shrink)

This text examines the dominant ways of looking at patient/clinician relationships in healthcare. By challenging these dominant views the author can explore presuppositions that are defective. She further explains how they come to be so readily and uncritically held and reinforced; and, why their implications can have such a profound affect on how we think and act. Using the methodology of philosopher, John Dewey, the author proposes an alternative bio/psycho/social approach to understanding the patient/clinician relationship and for resolving increasingly common (...) bioethical issues that arise in healthcare settings. Boulding to describe a unified view of evolution that encompasses all levels of reality, from the cosmic or physical through the biological, ecological, and sociobiological to the sociocultural. It focuses less on systems or any particular entity than on the processes through which they evolve. There are, however, a number ofsubstantive differences in what their proponents take``structuralism'' to be. This paper discusses the possibility of finding an ethic of at least partial and perhaps ever-growing content in a world not that of moral strangers (where we have nothing except our desire to live freely to unite us) and one of moral friends (in which values, goals and ways of doing things are held in common). I argue that both the world of moral strangers which Engelhardt's world view would support, as the world of moral friends which is the one (...) Pellegrino seeks both are untenable and that furthermore both can lead to a similar state of affairs. I suggest a dynamic world of moral acquaintances in which different belief systems and ways of doing things can come to some broad agreements about some essential thing. This is made possible because although we do not share the intimate framework Pellegrino might suggest, yet we are united by a much broader framework than the one moral strangers share. (shrink)

Elaborates an ethic in which beneficence on a personal and communal level has moral force; proposes the idea of an interplay between compassion and reason to help address moral problems; and sketches the conditions necessary for a democratic approach to such problems.

This edited volume explores the previously underacknowledged 'pre-history' of mathematical structuralism, showing that structuralism has deep roots in the history of modern mathematics. The contributors explore this history along two distinct but interconnected dimensions. First, they reconsider the methodological contributions of major figures in the history of mathematics. Second, they re-examine a range of philosophical reflections from mathematically-inclinded philosophers like Russell, Carnap, and Quine, whose work led to profound conclusions about logical, epistemological, and metaphysic.

Guarded fragments of first-order logic were recently introduced by Andreka, van Benthem and Nemeti; they consist of relational first-order formulae whose quantifiers are appropriately relativized by atoms. These fragments are interesting because they extend in a natural way many propositional modal logics, because they have useful model-theoretic properties and especially because they are decidable classes that avoid the usual syntactic restrictions (on the arity of relation symbols, the quantifier pattern or the number of variables) of almost all other known decidable (...) fragments of first-order logic. Here, we investigate the computational complexity of these fragments. We prove that the satisfiability problems for the guarded fragment (GF) and the loosely guarded fragment (LGF) of first-order logic are complete for deterministic double exponential time. For the subfragments that have only a bounded number of variables or only relation symbols of bounded arity, satisfiability is EXPTIME-complete. We further establish a tree model property for both the guarded fragment and the loosely guarded fragment, and give a proof of the finite model property of the guarded fragment. It is also shown that some natural, modest extensions of the guarded fragments are undecidable. (shrink)

We identify the computational complexity of the satisfiability problem for FO 2 , the fragment of first-order logic consisting of all relational first-order sentences with at most two distinct variables. Although this fragment was shown to be decidable a long time ago, the computational complexity of its decision problem has not been pinpointed so far. In 1975 Mortimer proved that FO 2 has the finite-model property, which means that if an FO 2 -sentence is satisfiable, then it has a finite (...) model. Moreover, Mortimer showed that every satisfiable FO 2 -sentence has a model whose size is at most doubly exponential in the size of the sentence. In this paper, we improve Mortimer's bound by one exponential and show that every satisfiable FO 2 -sentence has a model whose size is at most exponential in the size of the sentence. As a consequence, we establish that the satisfiability problem for FO 2 is NEXPTIME-complete. (shrink)
